Report from Business trip, Sweden, Bořík

I visited the Department of Chemistry of Umea University from 3 August to 3 September 2021. The aim of my training activity was acquaintance with the methods and instrumentation used for non-targeted metabolomics. Non-targeted analytical methods are fastly rising field in environmental research. These approaches are closely related to activities and projects running in our laboratory. During my stay, I had an opportunity to collaborate with associate professor Jerker Fick and Annika Johansson from The Swedish Metabolomics Centre. The training activity supported with program Erasmus+ brought me an overview of instrumentation, software, workflows, and quality control applied in another laboratory for non-targeted screening.

The internship was fixed for 3 months at the Lund University, Department of Immunotechnology, LUND, under the supervision of Dr. Fredrik Levander. The internship was aimed at gaining experience in routine molecular techniques as well as advanced proteomics techniques. Initially, I worked on routine tasks and research projects under supervision in the laboratory. I instructed in various methods.

  • Sample preparation (protein extraction, protein measurement)
  • Various Mass spectrometry-based techniques
  • Proteomics data analysis
  • Learning different bioinformatic tools

I processed samples designed for three independent proteomics experiments and produced data for publication. In short, experiment number 1- investigates the proteomics changes during nerve regeneration, 2- investigates the proteomics changes in locomotory organ regeneration and 3-Study the gender differences of eye proteome in crayfish.

My internship at ICM-CSIC was an inspiring journey enriched by a vibrant research environment. The institute's welcoming spirit was a remarkable experience. Learning new technology applied in animal behavior research provided priceless experiences during my stay at the ICM-CSIC. Acquiring advanced expertise in EthoVision XT, a challenging program for examining the behavior of aquatic animals (Fish and Crustaceans), was one of the highlights of my stay in Barcelona, Spain. This skill has opened new horizons in my understanding of animal behavior.

I gained a deeper understanding of academic concepts by participating in the seminars offered at the ICM-CSIC complex. Additionally, I'm happy to convey that our collaboration with the Renewable Marine Resources laboratory at ICM-CSIC was successful. I learned the matrix system  (El Temps and idTracker) software used for circadian rhythm analysis, paving the way for ground-breaking knowledge in these important areas.

I would like to express my sincere appreciation to my laboratory (Laboratory of Freshwater Ecosystems), our faculty (FFPW-RIFCH), and CENAKVA for providing me with this priceless opportunity. The experiences and knowledge gained during the three-month internship have undoubtedly set the course for my future endeavors.

I have completed a three-month traineeship at the Biotechnical Faculty, University of Ljubljana (BF-UL) from 9th February to 12th May 2023. The internship aimed to gain experience in advanced biotechnical and proteomics techniques. During my stay at BF-UL, I learned several methods for microbial protease activity characterization (phenotypic screening, e.g., “clear zones on agar plates”, azocasein method) and molecular (PCR amplification of selected genes) and correlation of the results collected for selected strains. These skills were applied to evaluate fish or fish products' quality and shelf life in a Food Model (homogenate of common carp meat), and the results were evaluated with professional software. I would like to thank the South Bohemia University project and ERASMUS+ grant for funding my stay in Slovenia.

Between November 12-26, 2021, I conducted a business trip to the IMR - Institute of Marine Research in Bergen (Havforskningsinstituttet), where I worked for the Virgin salmon Project as a postdoc researcher at the Department of Reproduction and Development from 2017 to 2020.  The Crispant dnd KO broodstock yet fertile (F0) I created for the Virgin salmon project has finally reached the breeding stage; therefore, germ cells (eggs, sperm) have been sampled for mutational and quality analyses. Also, I fertilized the best parents according to their genotypes. I microinjected dnd mRNA into the fertilized group (F1) to rescue germ cells which carry the sterility trait as dndloss of function mutation. After all the experiments, we evaluated the recent data and upcoming results to be able to present the second paper from this project. We drew a work plan for the further steps and paper submission deadline. All the current results of the project were presented at the department group meeting on November 25th. We have also discussed a new project idea to continue the collaboration and submit the proposal in February 2022 to the Norwegian grant.
